Effects of promethazine on nocturnal sleep in normal man. The effects of a phenothiazine, on sleep in ten healthy volunteers were investigated in two double blind polygraphic studies. The first part consisted of a single dose study with promethazine 50, 100, and 200 mg, using pentobarbital 100 mg as a reference substance. In the second part, four subjects spent 20 consecutive nights with nine drug nights (promethazine 100 mg), followed by a placebo withdrawal period of six nights, in the sleep (...) laboratory. Promethazine showed a dose related REM-depressing effect with a greater decrease, the higher the dose. The placebo value was 20.7% REM of total sleep time and gave 16.3, 13.5 and 11.4 respectively, for promethazine 50, 100 and 200 mg. Pentobarbital gave 16.2% REM. There was also an increase of stage II and with the highest dose an increase of stage III + IV. An increase of REM-latency together with a decrease of REM-periods was also seen, and while pentobarbital gave a decrease in REM-density

[On the respiration analeptic effect of fominoben HCl after administration of pethidine/promethazine (author's transl)]. The respiratory stimulatory effect of 3'chloro-2'-(N-methyl-N-[(morpholino-carbonyl)methyl]-aminomethyl)benzanilide hydrochloride (fominoben, Noleptan, PB 89) on the respiratory-depressing effect of pethidine/promethazine was investigated. In addition a double blind study was carried out on 36 patients with fominoben or placebo after pethidine/promethazine. To evaluate (...) respiratory function the vital capacity, Tiffeneau-value, volume of inhaled air/s, maximum breathing capacity, as well as the blood gas values (pO2, pCO2 and pH according to Astrup) were measured. The objective of the study was to assess whether ondansetron has superior nausea reduction compared with metoclopramide, promethazine, or saline placebo in emergency department (ED) adults.This randomized, placebo-controlled, double-blinded superiority trial was intended to enroll a convenience sample of 600 patients. Fixed drug eruption caused by three unrelated drugs: promethazine, pethidine and omeprazole. We report a case of fixed drug eruption (FDE) caused by three unrelated drugs. A 52-year-old woman presented with a diagnosis of FDE after taking a drug to treat a common cold. An oral provocation test and a patch test were carried out, and the patient reacted to promethazine, which is a derivative of phenothiazine. To determine the core structure of the antigen, we performed patch testing (...) and/or an oral provocation testing with six drugs that are similar in structure to promethazine. The patient reacted to five of the six drugs, and the antigenic determinant was identified as a phenothiazine and a tricyclic structure. The patient had similar eruptions at the same sites 3 years later after taking pethidine. Seven months after that reaction, a more severe eruption was caused by oral omeprazole. This is an extremely rare case of FDE caused by three structurally unrelated drugs.© The Author(s
